Probe description

The LSI Cyclin D1 FISH kit is intended for the determination of Cyclin D1 gene amplification in human tissues using fluorescence in situ hybridization (FISH).
The Cyclin D1 FISH kit contains a directly labeled fluorescent DNA probe in hybridization buffer. The fluorochrome Orange labeled Cyclin D1 probe covers the chromosome 11q13 region. The protein encoded by Cyclin D1 (CCND1) gene belongs to the highly conserved cyclin family. Cyclins and cyclin-dependent kinases (CDK) represent  the main regulatory systems of the cell cycle. The synthesis of Cyclin D1 is initiated during G1 and drives through the G1/S phase transition. Gene amplification or an increased production of the Cyclin D1 protein can be found in the different types of cancer, e. g. primary breast carcinoma, ovarian carcinoma, hepatocellular carcinoma, and small-cell lung carcinoma.

FISH results

A normal cell hybridized with the LSI Cyclin D1 (Orange)/CEP11 (Green) shows 2 orange signals of the Cyclin D1 gene and 2 green signals of chromosome 11. An abnormal cell hybridized with the LSI Cyclin D1 (Orange) shows more than 20 orange signals which indicate the amplification of Cyclin D1 gene.
